چکیده

The aim of this work was to evaluate the variability of the as1-casein locus in the endangered Girgentana dairy goat breed in order to define genetic improvement and a conservation program for this breed. The study was performed on 200 dairy goats by means of different PCR protocols. Themost frequent alleles were A (0.590) and F (0.290) followed by B (0.065) and N (0.047).CSN1S1 E allele was identified with a very low frequency (0.008). The most common genotype was AF (0.365) followed by AA (0.340). The high frequency of the strong genotypes is associated with the production of milk with high fat and protein content andwith optimal technological properties. In Girgentana goat breed, theCSN1S1 genotype information could be utilised in selection strategies formilk protein content andmilk yield, in order to select genetic lines for the production of ‘drinking milk’ using weak and null genotypes, and for niche products using strong genotypes. Additional keywords: Girgentana goat, milk production. Received 13 July 2012, accepted 13 October 2012, published online 7 February 2013

Genetic Polymorphism Detection of the Exon 1 Region of Acetyl-CoA Carboxylase Alpha Gene in Iranian Mahabadi Goat Breed

Acetyl-coenzyme A carboxylase α (ACC-alpha) is considered as the key regulatory enzyme in fatty acid biosynthesis. ACC-alpha gene is located on Caprine chromosome 11 and is polymorphic in many goat breeds. In the current study, we aimed to find possible single nucleotide polymorphisms (SNPs) in the exon 1 region of the ACC-alpha gene in Iranian Mahabadi goat breed. Genomic DNA was extracted fro...

Identification of the CSN1S1 allele in Indian goats by the PCR-RFLP method.

The allelic distributions of the CSN1S1 (αs1-casein) in the Indian goats are quite different from European goat breeds. Majority of Indian goat breeds and non-descript goats carry A, B, E and F alleles at αs1-casein locus, as found by analysing both DNA and protein levels. However, A and B alleles, known to be associated with better casein yield, were observed in the highest proportion in all t...

Genetic polymorphism at the CSN1S1 gene in two Czech goat breeds

The genetic polymorphism of the CSN1S1 (casein alpha-S1) locus was investigated in two endangered Czech goat breeds (White Shorthair and Brown Shorthair). These breeds are kept mainly for their good dairy performance. Genetic characterization of the CSN1S1 locus contributes to the knowledge of the genetic structure of these two endangered breeds. The study was performed on 498 goats (333 White ...

Casein haplotype structure in five Italian goat breeds.

The aim of this work was to investigate the genetic structure of the casein gene cluster in 5 Italian goat breeds and to evaluate the haplotype variability within and among populations. A total of 430 goats from Vallesana, Roccaverano, Jonica, Garganica, and Maltese breeds were genotyped at alphas1-casein (CSN1S1), alphas2-casein, (CSN1S2), beta-casein (CSN2), and kappa-casein (CSN3) loci using...

Short communication: genetic variability in the predicted microRNA target sites of caprine casein genes.

The main goal of the current work was to identify single nucleotide polymorphisms (SNP) that might create or disrupt microRNA (miRNA) target sites in the caprine casein genes. The 3' untranslated regions of the goat alpha(S1)-, alpha(S2)-, beta-, and kappa-casein genes (CSN1S1, CSN1S2, CSN2, and CSN3, respectively) were resequenced in 25 individuals of the Murciano-Granadina, Cashmere, Canarian...
